Mechanism

TracrRNA-crRNA directs Cas9’s action when fused as a single guide RNA (ie, sgRNA). For the target gene, an artificially designed sgRNA (small guide RNA, sgRNA) is used to identify the target gene sequence, and Cas9 protease is led to effectively cut the double-strand DNA to form the DNA double-strand break. Under normal circumstances, cells will repair the broken DNA through the method of efficient Non-homologous End Joining (NHEJ). However, during the repair process, mismatches including base insertion or deletion usually occur, resulting in frameshift mutations, which make the target gene lose its function, thereby achieving gene knockout.
